Evolution of regio-specific nucleoside kinases.

We have evolved RNA enzymes that turn a non-ligatable RNA substrate into a ligatable substrate by 5' phosphorylation. Previous in vitro evolution experiments have yielded both 2' and 5' kinase ribozymes. Our strategy makes use of a ligation-based strategy, that REQUIRES the evolving ribozymes to be regiospecific.  The substrate for phosphorylation is a tethered nucleoside that acts as a quasi-diffusible substrate, favoring the evolution of RNA enzymes that act may act on freely diffusing small metabolites.
